Effects of Antiviral Chemical on Eradication of Potato Virus S in Potato (Solanum tuberosum L.) Shoot - tip Culture

Abstract
This study was conducted to investigate the effect of antiviral chemicals on the eradication of potato virus S (PVS) from virus-infected potato (cv. Norchip and Red Pontiac) during in vitro culture. Among three antiviral chemicals used, acyclovir (ACV) and azidothymidine (AZT) had no eradicative effect on PVS in all the levels (0, 10, 20, 30 ppm), but ribavirin exhibited a significant eradicative effect on PVS in higher than 20 ppm. However, the highest level, 30 ppm, showed the undesirable effect on growth of potato shoots and microtubers. It was not possible to completely remove PVS of in vitro potato shoots by only one treatment of ribavirin in both cultivars. Therefore, the in vitro shoot tips infected by PVS should be repeatedly suhcultured with 20 ppm ribavirin media until no virus was detected. In cv. Norchip, it took 3 subcultures (months) to eradicate PVS from infected shoot tips, but in cv. Red Pontiac, it was not until about 7 subcultures (months) that PVS was no longer detected by ELISA test.
